Podtyp: Příspěvek ve sborníku (mimo kategorie RIV); In vitro growth-inhibitory effect of plant-derived compounds and their derivatives against diarrhea causing microorganisms. 2014, Materials and Methods The compounds tested (Tab. 1) were purchased from Sigma-Aldrich (CZ) and Chromadex (USA). All bacterial strains (Tab. 1) were obtained from Oxoid (UK). The minimum inhibitory concentrations (MICs) were determined by the broth microdilution method [1] using 96-well microtitre plates. The growth turbidity was measured by Multiscan Ascent Microplate Reader (Thermo Fisher Scientific, USA) at 405 nm. MICs were calculated based on the density of the growth control and were expressed as the lowest extract concentrations that resulted in 80% growth reduction compared to that of the compound-free growth control [4]. Results and Discussion The best results (Tab 1) were obtained for zinc pyrithione with the MIC 4 µg/mL followed by sanguinarine with MICs ranking from 16 to 32µg/mL. Moderate antibacterial effect has been observed for rhein and pterostilbene on Enterococcus faecalis with the MICs 32 and 64 µg/mL, respectively. Zinc pyrithione is a compound used in treating dandruff and seborrhoeic dermatitis [5], [6]. According to our best knowledge, there are no reports on antibacterial effect of zinc pyrithione, a compound used in treating dandruff and seborrhoeic dermatitis [5], on representatives of diarrhea causing microorganisms. Zinc pyrithione just show teratogenic effect seen in the study of Nolen and Dierckman 1979, where the increased incidences of ribs defects in the fetuses of rats given zinc pyrithione orally at level of 15mg/kg body weight. Sanguinarine is an alkaloid found in many medicinal plants. Sanguinarine has cytotoxic or antiproliferative effects on several normal or cancer cell lines, including human gingival ?broblasts (Malikova et al., 2006b), rat hepatocytes (Choy et al., 2008), human promyelocytic leukemia HL-60 (Vrba et al., 2009), and human osteosarcoma cells (Park et al., 2010) Conclusions Pyrithion (Zinc pyrithion) is a compound commonly used as a drug in the treatment of diarrheal diseases [5], possessed strongest antidiarrheal activity among all substances tested in this study. According to our best knowledge, this is the first report of antibacterial properties of this compound. .
