The pregnancy diagnosis in half-tamed animals is difficult. The aim of this research was to verify the Cuboni reaction and barium chloride test in Bactrian camels (Camelus bactrianus) and their usage as a non-invasive tool for diagnosis of pregnancy from the urine. The urine of nine adult female camels from Prague, Plzen and Bratislava zoological gardens was collected to the half-litre plastic cups fasted to 1.5 meters long rod in one or two months intervals. Forty one samples were evaluated during September 2010 to February 2011. The samples of urine were stored in a refrigerator and the analyses were carried out within 14 days after sampling. The Cuboni reaction was performed by the State Veterinary Institute Prague. The minimal amount of the sample was 20 ml and the reaction was evaluated like positive, negative, and dubious. The barium chloride test was done in a laboratory of the university by 1% solution of barium chloride. One, five, ten, and fifteen drops of solution were added to the 5 ml of urine and the reactions (presence of white precipitate) were described. Both tests are based on chemical reaction of oestrogen in urine. Three females were pregnant and give a birth later. The Cuboni test of urine in these females showed in 86.7% positive and in 13.3% dubious reactions. The positive reaction was noted even 215 days before parturition. We did not find any false negative reaction but we noted positive and dubious results in non-pregnant females as well. There is high probability that camel urine reacts positive even if the female is in the heat due to higher oestrogen levels than in mares of domestic horses. We did not find any clear relationship between the reproductive status of females and the results of barium chloride tests. There was also no correlation between results of Cuboni and barium chloride reactions. The Cuboni test can be a useful tool for pregnancy diagnosis in camels during the second half of gestation. If the urine capture is repeated 10-14 days after first sampling, the false positive result, due to the possibility of the heat, can be eliminated. The research of Cuboni tests will continue and will be more focussed on the first half of gestation, and the time when the first positive reaction of urine will occur. For consumers one of the most important one is its toughness, but this characteristic for game meat is often scored lower when cooked. Popularity of consumption of raw tartare is increasing among the Czech consumers. Therefore, we focused on evaluation of texture by Warner-Bratzler shear force (Instron analyser) on raw samples of eland meat to find out the best cuts. Eland bulls were slaughtered at age of 1.5 (n=6) and 3 years (n=11). Elands were captive bred, finished by neck shot at the farm and processed in slaughter house. During dissection 24 hours after the slaughter from chilled carcass we collect samples from 12 muscles. The highest values of shear force, which represents the most tough muscles, had m. sternomandibularis (395±12 N, LSmean±SE) and pectoralis profundus (227±11 N). The lowest values had m. longissimus lumborum (43±11 N), psoas major (60±13 N) and m. sartorius (67±11 N). Values below 100 N were measured also in m. semitendinosus, biceps femoris and gluteus medius. Muscles triceps brachii, quadriceps femoris, m. infraspinatus and flexor digitorum had values between 100 – 200 N. We had not found differences in shear values between samples from 1.5 and 3 years old animals during comparison of the same muscles. Abstract: Laboratory equipment for sampling is often expensive and sometimes cannot be used for animals. We needed to collect samples of saliva and urine from even-toed ungulates, concretely from Bactrian camels (Camelus bactrianus) kept in zoological gardens and domestic cattle (Holstein dairy cows), for the research focused on reproduction which was realised between April 2010 and February 2012. So we tried to find cheap supplies for sampling. The sampling of urine by plastic cup mounted to rod is quite common for urine sampling. We used clear plastic PET cups with volume of 0.5 litre mounted by wire to broom handle for urine collection from females camels. When the female started to urinate, we approached toward her and collected the urine to the cup. We collected by this way 78 samples of urine from 13 female camels. Most of animals had no problem with the collection. Only three females (16.6 %) were “shy” and stopped urination when someone got closer to her. But it was usually possible to collect urine from these females as well. The sampling of small amount of saliva was realised by plastic coffee stirrers. Saliva production was stimulated by dry bread in case of camels or by the hand inside the mouth in case of cattle. We collected successfully 623 saliva samples from 11 camels and 408 saliva samples from 8 cows. We also collected saliva of camels in bigger amount for hormonal assays. We used the bigger plastic spoon for sampling. The handle of the spoon was plunged to the mouth when a camel was trying to attempt the bread and saliva got stuck in the depression of the spoon handle. Saliva was removed by coffee stirrer to test tube and the process was repeated several times until we collected more than 2 ml of foamy saliva. We collected successfully 422 samples from 16 camels. Camels behaved to be fed from hand had no problem with the collection. We concluded that plastic catering supplies are adequate and cheap equipment for sampling. Abstract: There is an increased requirement for cheap and effective detection methods for safety and quality control in production animals. RnAssays has made possible the detection of bacterial, viral and/or parasitic infections and chemical contaminants simultaneously in a single test portion in a single analysis run, using flow cytometry. These tests allow for health profiles of herds to be established, as well as detecting transmissible diseases (zoonoses), residues of veterinary antibiotics at slaughter, simultaneous detection and chemical contaminants (Jankovska I., Miholova D. and Lukesova D. et al., 2012). RnAssays produces screening assays for a wide range of matrices including plasma, serum, eggs, and meat (drip) principally from pigs, poultry and bovine animals. A combination of fluorescent beads and a unique bead surface modification means that both chemical contaminants (e.g. antibiotic residues) and microbial contaminants (e.g. viral and bacterial pathogens and parasites) can be detected at the same time from the same sample. The combination of bead sizes with different fluorescent intensities creates powerful and flexible multiplex possibilities (Aldert A. Bergwerff, 2012). In 2010 the World Health Organisation estimated that every year, 2.2 million people around the world die from food infections, the majority of them children. Zoonoses represent one possible source of such infections (Lukesova et al., 1998). RnAssays range of Zoo Plex™ serology tests help food producers monitor animal herds to confirm the ongoing absence of zoonoses such as Salmonella, Toxoplasma, Trichinella, Taenia (cysticercosis) and others (Dubey, 2010). The SPF (Specific Pathogen Free) system introduced by Denmark is aimed at protecting pig herds from infection, limiting and preventing the spread of infection outbreaks and setting standards for animal transport. RnAssays is developing serological SPF Plex™ assays to meet this requirement for providing herd profiles with respect to animal diseases in a single multiplexed test. The Residue Plex™ assay uses monoclonal and polyclonal antibodies and natural capture agents in a competitive inhibition assay configuration. Discrimination of the individual microspheres is arranged by different bead sizes and bead internal fluorescent intensities, which lead to the analysis of multiple analytic results per sample. This new bead-based test, which detects e.g. T. gondii and T. spiralis antibodies simultaneously within each sample, can replace two indirect tests for the determination of respective antibodies separately, while performing equally well or better. This will bring significant savings for its customers in terms of financial resources and time and will deliver improved product safety. One of the advantages of a bead-based assay over ELISA is the possibility to determine multiple specific antibody responses per single sample run facilitated by a series of antigens coupled to identifiable bead-levels. In conclusion, this initial study of a novel bead-based assay capable of a simultaneous detection of serological antibodies against T. gondii and T. spiralis antigens indicates that the test results correspond very well to the infection status of the animals, and, furthermore, there is a substantial to excellent agreement with other indirect tests (Gertie CAM Bolkem, Aldert A Bergwerff and Frans Knapen, 2012). In order to estimate the applicability of this test for purposes of serological monitoring, further testing of sera from naturally infected animals is required. RnAssays tests are designed for the highest possible throughput with minimal manual intervention to allow the processing of a maximum of over 1900 samples per day..

Feed Consumption, Carcass Evaluation and Growth Performance of Broiler Rabbits Fed Different Levels and Processing Methods of Milk Thistle (Silybum marianum) Supplement. 2016, Milk thistle (Silybum marianum) belongs to the family Asteraceae which are widely spread in arid and semi-arid areas of Mediterranean regions. This annual plant contains silymarin-flavolignans with hepatoprotective and canceroprotective properties which show positive effects on health and performance of animals. The study aimed to investigate the effect of different level and processing methods of milk thistle (Silybum marianum) on feed consumption, carcass composition and growth performance of broiler rabbits. A total of 180 HYLA broiler rabbits, 42 days old, were fed with different concentrations of milk thistle supplement - group III (0.5 % of fermented milk thistle) and group II (1% non-fermented milk thistle) in comparison with control group I (standard feed ration without any supplementation). Feed and water were available ad libitum. The experiment started at 42 days of rabbit age and finished by slaughter when rabbits achieved 2.6 kg of live weight. The weight of the body parts, carcass weight, carcass yield, and growth performance were recorded and calculated. Carcass weight and carcass yield in rabbits fed with 0.5 % fermented milk thistle were significantly higher (P<0.05) than in rabbits of other groups. However, there were no significant differences (P>0.05) between control and experimental groups in the growth performance. Furthermore, daily feed consumption was higher in the treatment group II and group III compared to Control I (P<0.05). The results of this experiment therefore indicated that 1% non-fermented milk thistle extract supplemented in the feed ration for broiler rabbits is not a suitable supplement for improvement of broiler rabbits´ performance. However, 0.5% fermented milk thistle could be used to improve the carcass performance. .

Sensory Evaluation of Liver/Meat Pates Made from Fresh or Frozen Meat.. 2018, Small animal producers in Europe can financially benefit more from sale of value added product then from only carcasses or dissected cuts. Pâté is a traditional and popular product, which enables to process low value meat cuts and edible offal. Especially, when introducing on market a new exotic species like eland (Taurotragus oryx), sale of low valued cuts with higher collagen content is problematic. Therefore, we aimed to evaluate sensory traits of several pates made of liver and less valuable fresh or frozen meat cuts (neck, brisket and plate) of eland and cattle. Four batches of pâté made of fresh and frozen material (45 and 90 days) was tested by 35 panellists in 14 organoleptic traits. Two pâté batches consisted of chicken liver and beef or eland meat. Another two were made from eland or beef liver together with eland meat. Pâté made from fresh material including chicken or eland or beef liver together with eland meat or beef has showed mainly only differences in textural characteristics like in friability, overall texture being better scored for pâté including chicken livers. Chicken livers resulted also in higher intensity of colour. Freezing of material (meat and livers) before processing into pate resulted in worse scoring of final product being better scored for batches with chicken livers for sensory traits of overall appearance; pleasantness of taste, colour and intensity of colour; colour hue and textural friability; overall texture and of pleasantness of consistency. Bovine cysticercosis as a zoonotic disease is spread in different regions across the globe and which affects both people and cattle. It is caused by the tapeworm Taenia saginata (T. saginata), whose final hosts are humans while cattle are the most common intermediate host. The cattle are infected by swallowing tapeworm eggs from human feces from vegetation in the environment or water often after flooding and they can survive in the environment for several weeks or months. After being ingested by cattle, they evolve and turn into larvae and afterwards migrate to muscle tissue (mainly to heart, m. masseter, diaphragm or tongue.
